GLIO-1

GLIO-1 is an orally-delivered, highly potent and brain penetrant inhibitor of the enzyme DHODH. It was invented by the same team who discovered vidofludimus calcium, Immunic’s drug candidate currently completing phase 3 clinical trials in multiple sclerosis.  Vidofludimus calcium has shown potent inhibition of DHODH in preclinical testing and also appears to have a favorable safety and tolerability profile in clinical trials in multiple sclerosis patients and other auto-immune conditions, to date. Vidofludimus calcium is an investigational compound and not approved for use in any indication.

GLIO-1 shares vidofludimus calcium’s chemical backbone, but also contains key differences designed for neuro-oncology, including both heightened potency and target selectivity. 

In preclinical testing, GLIO-1 has shown:

  • Highly potent inhibition of the human DHODH enzyme
  • Strong brain penetrance in animal models
  • Potent cytotoxicity in human brain tumor cells with sparing of wild-type cells
  • Statistically significant improvement in survival in gold-standard patient-derived xenograft models
  • Clean tolerability in animals with chronic daily dosing (out to 120 days)
